For me it would be
Legend of Zelda Ocarina of Time - This might be the first zelda game i ever played (i may have played link to the past at my grandpa's house before this). When my friend showed it to me I was hooked. I played it for days because it was so fun. And when my otherfriend played it we became the biggest zelda fans in the school.
castlevania circle of the moon - this is because it was the first castlevania game i ever played. I can remember bringing my gameboy advance on a camping trip and my cousin bringing his. When i borrowed the game from him after seeing how he played it at christmas i wouldnt stop playing until the rechargeable battery i bought for it died then i would go outside and play in the woods and then go baack and play it when it was recharged.

I too have to go with Ocarina of Time because it brought me and my friends together every rainy day. That and the original Smash Bros.
Also, I'm pretty sure Pokemon Silver as my first Pokemon Game and Duck Hunt as my first game at all.

Mega Man X -- I played this more than any other SNES game I've played. I know where to find EVERYTHING in the game -- all the power-ups, hear tanks, etc. I could go on and on about the music if I wanted to, but that would almost be off-topic.
Super Mario Bros. 3 -- I actually enjoyed all the other Mario games when I was a youngster, but this was always my favorite. Still is to this day, actually. Whenever I hear the happy-sounding music of the first world, I can't help but feel happy.
Contra 3: The Alien Wars -- I actually prefer the original Contra game, but this is the first I ever played in the series. A friend of mine brought it over to my house and man I didn't want him to take it back with him! Haha. Such a fun game.
Resident Evil 2 -- I didn't have a PS1 when I was younger, and I was glad when this got released on the N64. It was the first survival horror game I played, and while it's not the scariest for me, it is certainly the most memorable. The musical score, the storyline, the gameplay, the monsters, and the atmosphere are some of the greatest factors of the game... and that's probably not all. I could write an essay on this game.
Much later in the future, Guitar Hero 2 might be another game that I consider special to me.

Twisted Metal 2: My brother and I would spend countless hours playing co-op and coming up with new ways to play. Whenever I go home we still put it in and play it.
Crash Bash: Another one we had a lot of fun playing together that we still put in and play.
Donkey Kong Country 2: This one I used to play with a friend of mine in 4th grade. The music and the environments were so amazing the first time I played. Bramble Scramble is still one of the most breath taking pieces of music I've ever heard in a game.
Mystical Ninja 64- This game is what made me buy a Nintendo 64, like DKC2 I loved the music and environments. The gameplay is still fun too, unlike DKC2 which hasn't aged well gameplay wise. One of my favorite games of all time.

Well, divided by system:
Nes-I would say 8 eyes, final fantasy, goonies 2, and castlevania. I either inherited these games or found them in pawn shops or thrift stores, leaving them without box or manual and thus without context, where I stepped in to fill the blanks. All of them either hit on latent tastes or laid the groundwork for their formation, that is my love of exploration, the fantasy genre, gothic horror elements, and a sense of the unknown. Each one has powerful nostalgia for me and as said before probably had a strong effect my tastes to this day.
SNES-LoZ LttP, FF6, and Super metroid. Not the most obscure selections, but ones that are very close to my heart. I was poor growing up, I got my SNES at the end of its lifespan and so these are the games that laid beyond my reach. Until I bought my own system I saw fleeting glimpses of them, a manual here and there or a quick play at the house of a friend of a friend. My heart yearned for them for many years, and when I finally got to play them they did not disappoint. Scenes like the dying metroid or shadow parting with interceptor have more emotional resonance with me than movies like old yeller, and legend of zelda says adventure more to me than lord of the rings.
The big one above all-SFII. This is my game. It was the first game I ever played, it was with my dad at an Allsups and I was playing as Sagat against the computer Ryu. I was too young to use anything but the roundhouse kick and as was par for the course in the day had my ass summarily handed to me. But from then on I was hooked. Whenever I went to an arcade, SFII was my base choice cause every arcade, laudromat, and theater had have one. I took sides against mortal combat because of it, and found my choice vindicated by the loser having a shitty fight system and a draw based entirely on shock novelty. It turned me on to good versus fighters, to martial arts movies, to Capcom, and maybe videogames in general.
I might like videogames too much.

Final Fantasy 6--it was my first FF game. The soundtrack really brings me back to my days of having entire days of just sitting and playing it (with obvious breaks for the bathroom and food). Considering what's been done with IV as a DS game, I'm still hoping Squenix gets around to a VI DS.
Secret of Mana--the only RPG I can remember scrounging for pennies to meet enough coin to rent it for "just one more day" from MicroPlay.
Xenosaga--It's somewhat hard to find people that have played the series, with most of my inqueries being met with "You mean Xenogears, right?" or "Well, I played Xenogears." I bought the first 2 games used when I bought my used PS2 and got Episode 3 the first day it came out.
Hunter: the Reckoning--So many accidental friend kills with the rocket launcher...

Well, the one that instantly comes to mind is the original Final Fantasy. I honestly think that flying around in the airship for the first time, coupled with finally, finally, FINALLY achieving the class change after god knows how many hours of grinding, and tons of attempts at the Castle of Ordeal, is the highlight of my gaming life. Nothing since then has given me nearly as much satisfaction or elation as a player.
The first two Legend of Zeldas, especially (oddly enough) AoL. I loved the first one, don't get me wrong, but that one was FUN as all hell. The second one was damn near punishment to play, and yet I kept coming back for more. We have something of a masochistic relationship, AoL and I, but we can't live without each other either.
Those were my obvious choices. Now for the weird-ass ones, both courtesy the PC (or rather DOS, which was what passed the for PC gaming then...)
Comanche: Maximum Overkill: Fuck. Yeah. Bitches. This was the greatest and most exciting game of my very young life. I blew the shit out of more Russian tanks and helicopters than I have eaten meals. And those sound effects...and of course the VoxBox graphics. God I loved that game. To this day, I still perfectly remember the command I had to type in on my dad's old DOS. And of course, that THX-like sound cue when the NovaLogic logo came up...
WolfPack: Worst sub sim ever to some people, dear memory of hundreds of hours to me. In spite of my patriotism I could never bring myself to play the Americans...I mean who wants to be stuck in a destroyer when you could be commanding a damn U-Boat fleet?! This game was so damn fun. It was my first brush with designing my own mission too, thanks to its primitive mission editor.
Flight Simulator: The original one. God I loved this game. Designed my own planes, kept a flight log, made up elaborate missions for flying from one airport to another... This one was especially dear to me because I wanted to be a pilot more than anything else in the world. Well I still do, but my height and bad eyes have nixed that. I still remember this game fondly as the closest I'll ever come to that, though. Damn that was good times. Making it all the way from O'Hare airport to JFK in real time is still one of my proudest moments.

I don't think anyone else played this game, or even remembers it, but Destruction Derby for the original PSone holds a special place in my heart and always will. I've probably put in more hours in mastering that game than any other and it was right in that sweet spot of teenager nostalgia that I'll always remember. Also, for some reason, I always played Alanis Morisette's Jagged Little Pill CD when I played the game, so I also have a weird musical association with it in that whenever I hear "Hand in my pocket", I think about Destruction Derby.
Also, I'll always remember playing through and finishing Final Fantasy 9 because it was the first FF game I actually completed, and I did so just before I moved to England on an epic adventure of my own. So that atmosphere of sitting in my bedroom, completing an epic game just before beginning my own unknown journey that will take me away from home for an indeterminate amount of time will always stick with me.

I remember Destruction Derby! My cousin had it, and it was awesome.
GoldenEye007 - One of the first shooters I ever played, and cemented my reputation on my block as the Lord of The Shooter.
Adyin Chronicles: The First Mage - Okay. No one knows this game, but I read about it in Nintendo Power one day and had to have it. Sadly, I never bought it, only rented on a regualr basis. Wasn't good, but damnit, I loved that RPG.
Mass Effect - The first game I bought for my 360, along with Saint's Row. I was on crutches that Christmas, so me and my cousin drove up to GameCrazy and bought me the game. I still have it.
Guitar Hero 2 - I loved rocking with my friends, and the bass part actually convinced me to start playing my bass again.

didn't want to do it, because it's not cool to like ff7 anymore, but that game will hold a special place in my heart. first game to keep me up till 4 in the morning only to wake up and pick up where I left off at like 7 or 8am.
But before that probably simons quest as it is the first game that kind of creeped me out as well as zelda for snes.
Oh and not anything special about the game, but smb was the first game I played, and it brings back fond memories of going of to my neighbor/girlfriends house to go swimming and play mario. yeah she was pretty much my girlfriend out of necessity...
